Chtop (5E6) Rat Monoclonal Antibody

IHC-P analysis of mouse spleen tissue by anti-mouse Chtop antibody (AP93630). IHC-P was performed using sections of the formalin-fixed paraffin-embedded mouse spleen tissue. Antigen was retrieved through addition of boiling Tris/EDTA buffer pH 9 in a pressure cooker for 3 min. Endogenous peroxidase activity was quenched by incubating the sections with 3% H2O2 for 30 min at room temperature. The sections were then incubated with anti-mouse Chtop primary antibody (AP93630) at 5 µg/mL at room temperature for 1 h. Poly-peroxidase conjugated goat anti-mouse IgG (which cross reacts with rat IgG ) was used as the secondary antibody. Diaminobenzidine was used as the chromogen. The section was counterstained with hematoxylin. A tissue section incubated with phosphate-buffered saline followed by incubation with the secondary antibody was used as the background control. Result: Cells in red pulp and cells of white pulp are positively stained at the nuclei.

Function	Plays an important role in the ligand-dependent activation of estrogen receptor target genes (By similarity). May play a role in the silencing of fetal globin genes (PubMed:20688955). Recruits the 5FMC complex to ZNF148, leading to desumoylation of ZNF148 and subsequent transactivation of ZNF148 target genes (PubMed:22872859). Required for the tumorigenicity of glioblastoma cells. Binds to 5- hydroxymethylcytosine (5hmC) and associates with the methylosome complex containing PRMT1, PRMT5, MEP50 and ERH. The CHTOP-methylosome complex associated with 5hmC methylates H4R3 and transactivates genes involved in glioblastomagenesis (PubMed:25284789).
